TNFRSF13C(又称B-cell-activating factor receptor,BAFF-R)是与BAFF结合的特异性受体。BAFF-R作为B细胞谱系表面的标志物,在多种B细胞恶性肿瘤中高度表达,是免疫治疗的理想靶点。在机制上,BAFF-R信号激活NFkB通路,促进肿瘤存活和增殖。B细胞淋巴瘤和急性淋巴细胞白血病(acute lymphoblastic leukaemia,ALL)患者中BAFF-R表达增加与疾病进展相关。相关研究报道已表明,BAFF-R信号是B细胞生长和存活的驱动因素。这一关键特征提示,可以通过下调BAFF-R表达来限制B细胞肿瘤逃避治疗的能力。BAFF/BAFF-R轴已成功地用于自身免疫性疾病,特别是针对BAFF配体的单克隆抗体。最近,靶向BAFF-R的单克隆抗体在临床前模型中显示了对慢性淋巴细胞性白血病(CLL)的疗效,尤其是与Bruton酪氨酸激酶抑制剂联合使用时,该单克隆抗体已进入临床试验(NCT03400176)。
