CD33

CD33又名氨基肽酶N(APN),是一种跨膜糖蛋白,属于唾液酸结合免疫球蛋白样凝集素的一员。CD33在交联或者配体结合后激活,介导抑制信号,调节细胞内钙动员、细胞粘附、白血病细胞凋亡、髓系细胞成熟和细胞因子的产生等等。CD33在90%以上的急性髓系白血病患者都有表达。在造血干细胞表面没有表达,在成熟的粒细胞和其他组织也没有表达,CD33作为髓系白血病治疗的潜力靶点,一批企业相继布局了CD33药物的研发,辉瑞的靶向CD33的ADC药物Mylotarg已获批上市。除了ADC药物外,靶向CD33的双抗药物和CAR-T细胞疗法亦在如火如荼地研发中。
